研究概要	本研究の目的は、グルタミン酸受容体を分子レベルから解析する事により、グルタミン酸の受容機構と本受容体の高次脳神経機能における役割を明らかにすることである。初年度である平成3年度の成果をまとめると、1.既にクロ-ン化しているメタボトロピック・グルタミン酸受容体(mGluR1)を用い、新たに4種類のmGluR cDNA(mGluR2ーmGluR5)を単離することに成功した。mGluR1ーmGluR5は、7回の膜貫通部位の前に大きな細胞外ドメインを持つ、互いに高い相同性を示す構造を有し、mGluRは一群のG蛋白共役受容体群を形成している。2.mGluR1,R5,R2を動物細胞で発現し、前2者はIP_3/Ca^<2+>細胞内情報伝達系に、又後者は抑制性のcAMP系に共役していること、mGluR1/R5とmGluR2はアゴニストの選択性が異なることを示した。mGluRはIP_3/Ca^<2+>系のみに共役すると考えられており、この事実は、mGluRの機能を考える上で重要な問題を提起している。3.mGluR mRNAの発現は、相互に異なった発現部位を示す。4.グルタミン酸受容体の中で高次脳神経機能に中心的な役割を果たすNMDA受容体のクロ-ン化に成功し、本受容体は938個のアミノ酸からなり、4回の膜貫通部位を持つligandーgated ion channelであることを明らかにした。2.カエル卵母細胞で発現し、NMDA受容体は、特異的なアゴニスト、アンタゴニストの作用、Ca^<2+>透過性、膜電位依存性Mg^<2+>ブロック、グリシンによる活性化、Zn^<2+>による阻害等、本受容体に特徴的な性質を全て示す多機能受容体であることを明らかにした。3.本受容体mRNAは、中枢神経系のほぼ全ての神経細胞で発現している。
