巨大分子集合体プロテアソ-ムの構造と細胞生物学的機能に関する研究

研究概要

(1)cDNAクロ-ニングによる構造解析研究:本年度に新たに5種のプロテアソ-ム成分の一次構造を決定し、従来の成果と合わせた考察からプロテアソ-ムが進化的に保存された新しい遺伝子ファミリ-を形成していると言うプロテアソ-ムの分子進化に関する新概念を提唱した。
(2)遺伝子クロ-ニングと転写調節研究:ヒトプロテアソ-ムのHC3遺伝子の全構造を決定すると共に、5'ー上流領域の発現制御部位の解析から、転写を負に調節する部位を同定した。また、プロテアソ-ムを構成する多遺伝子群の協調発現機構を解明する為、HC2,HC5,HC8の遺伝子をクロ-ニングし、現在その全塩基配列を決定中である。
(3)プロテアソ-ム依存性の蛋白分解機構とユビキチンによる分子識別機構の解析:20Sプロテアソ-ムは複数の制御蛋白質群とエネルギ-依存的に会合して分子量200万・26Sの巨大な分子集合体を形成し、この蛋白質超分子複合体はユビキチンによって標識された不安定な細胞内蛋白質をATPの代謝エネルギ-を利用して分解する。本研究ではヒト及びラットの各組織からこの26S型プロテアソ-ムを大量に純化精製すると共に電子顕微鏡による解析から本酵素がダンベル型の分子形状をもつ巨大粒子であることを示唆した。またこの巨大複合体のサブユニット構造を明らかにすると共に現在、その主要成分のcDNAを単離中である。
(4)プロテアソ-ムと細胞増殖:我々は酵母プロテアソ-ムの遺伝子破壊が致死性を誘発することからプロテアソ-ムが細胞増殖に必須であることを証明すると共にヒトの各種癌細胞においてもプロテアソ-ムの異常発現を観察し、本酵素が高等生物の発生・成長及び癌化に密接に関係していることを示唆した。
(5)プロテアソ-ムと抗原プロセシング・提示:最近ヒトクラスII MHC遺伝子領域に二種のプロテアソ-ムのサブユニット遺伝子が存在することから、プロテアソ-ムの内在性抗原のプロセシングへの関与が示唆された。我々はプロテアソ-ムの全遺伝子群がインタ-フェロンーγによって強く誘導されることを発見したので、現在この酵素複合体が免疫応答遺伝子として作用する可能性について転写制御と細胞機能の両面から検討中である。
