アルツハイマ-神経原線維変化(PHF)内tauの翻訳後修飾の同定

研究概要	本年度はアルツハイマ-病脳に蓄積するPHFの構成成分tauの異常なリン酸化部位の同定を開始した。 1.間接的アプロ-チ(免疫化学的手法) まず特異性の異なる2種類の抗PHF抗体(SDSーPHFに対する抗体、ギ酸処理したPHFに対する抗体)を用いてPHF内tauのリン酸化部位の同定を試みた。2種類の抗体からアフィニティ-カラムによりリン酸化tau特異抗体(抗ptau1、抗patu2)を精製し、プロ-ブとした。抗ptau1、2は胎児型のリン酸化tauと反応する。そこでラットの胎児型tauを精製し、種々のプロテア-ゼ消化により得られたペプチドを逆相HPLCで分離し、抗原活性をELISAで検出した。さらに免疫原活性のあるフラグメントについてアミノ酸配列と質量分析を行った。その結果これら2つの抗体はAsp313ーIle328のリン酸化フラグメントと強く反応することが判明した。さらに抗ptau2の認識するエピト-プはSer315のリン酸化と同定された。このSerは既知のキナ-ゼのcanonical sequence上にはなく、未知のキナ-ゼによるものであると考えられる。 2.直接的アプロ-チ次にPHF内tauのリン酸化部位を蛋白化学的に同定するために、まずPHF内tauの精製法を確立した。アルツハイマ-病脳のSarkosyl不溶性画分をグアニジン塩酸で可溶化し、TSKG3000でゲル濾過を行い、tauを含む画分をセミマイクロボア逆相HPLCで精製した。この方法によりPHFtauを高純度に効率良く精製することができた。またこの過程でPHFtauと正常tauの両方にN末端側がプロセスされたtauが存在することが明らかになった。同時に、highly modified tauの画分、即ち、スメアを呈するPHF内tauの画分及びubiquitin化を受けていると思われるPHFの画分も得られることがわかった。
