研究概要	本研究班は生体や組織が病的状態あるいは特殊な環境下におかれたときに発癌のイニシエ-ションがいがに変動するかを知ることを目的とする。本年度の成果を以下に示す。()内は研究分担者名である。 1.DNA修復酵素遺伝子adaを導入したトランスジェニックマウスを樹立し、そのマウスでDNA修復と酵素活性の関係を解析した(石川)。 2.神経線維腫症のハムスタ-モデルを用いて、その初期病変を解析すると共にがん遺伝子neuの変異を明らかにした[中村(卓)]。 3.DNA誤まり修復遺伝子mucABをBALB3T3細胞に導入させたところ、細胞が癌化形質を示すことを見い出した(田の岡)。 4.各種発癌物質で修飾したDNA鋳型としてDNA合成を行なわせるとDNA体加体によってDNA合成発応が一定の影響を受けた(多田)。 5.マウス肝細胞の初代培養系でのチトクロ-ムP450 1A2の誘導発現には培地中のプロリレの存在が必要である(根本)。 6.家族性大腸線腫における正常粘膜部での細胞回転を調べたところ、健常人のそれと増殖帯を比較して異常は認めなかった[中村(真)]。 7.肝硬変症について生検標本を用いて肝装胞のS期細胞を計測した結果そのLIの高値群からより早く肝癌が発生することを認めた(清水)。 8.鉄による発癌は鉄を触媒するフリ-ラジカル反応が関与し骨癌を発生させる鉄ニトリロ三酢酵は強力なラジカルの発生源である(岡田)。 9.MNNGを用いた特定のプラズミドDNA上に突然変異を誘発させさせ、その変異を分折したところ、ホットスポットを認めた(真木)。 10.UV誘発マウス皮膚腫瘍についてras遺伝子の活性化を調べるとUV損傷とras突然変異との間に興味ある関係を見出した(錦織)。 11.ラット肝細胞の培養系においてニコチナマイド添加や培養波の改良により肝細胞の長期培養が可能となった(三高)。
