研究概要

劣性表現を持つ癌抑制遺伝子の機能の検証は細胞が持つ一対の相同染色体の両側において当該遺伝子を欠損させなければならないが、それを可能にしたのが、マウス培養胚幹細胞(ES細胞)における相同組換えである。これにより正常遺伝子を欠損導入遺伝子と取替えることができる。そして、この相同組換え胚細胞由来のマウスの交配により両側の当該遺伝子を欠損させたマウスを得、劣性表現を示す当該遺伝子の機能を探る方法の効率化を検討する。そして、ES細胞における相同組換えを利用して個々に特徴ある発癌に関連する遺伝子、特にがん抑制遺伝子の生物個体における機能を解析することを目的としたものである。
まず、キメラ形成能の高いES細胞系の樹立では、井川は、相沢らの協力で、正常3日胚(これまでは4日胚を用いた)からの樹立で、これに成功した。また、城石はMHC領域に高頻度組換えを起こすマウス系B10ーA(R209)のES細胞株ESR209.1を樹立しキメラ形成能を確認した。また、中辻はC57BL/6よりES細胞系を樹立し、その生殖系列キメラ形成能を確認した。また、卵原細胞を体外で培養し、雌マウスに移入し、操作された生殖細胞由来の子孫を得た。ES細胞系における相同組換えの高率化では、井川が、相沢らの協力で、ジフテリア毒素Aフラグメント遺伝子をネガティブ選択に用いることで成功し、実際に、ES細胞系でp53など3つの遺伝子の欠損導入を果たした。木村は、CーHaーrasに偽遺伝子がないことを確かめ、相同組換え用の同遺伝子を含むDNA断片をクロ-ン化した。高橋は、マウスHox3.5遺伝子を組換え用に構築した。若杉は、山村と共同で、相同組換えで欠損させるマウスNFー1遺伝子を単離した。また、野田(哲)は、マウスにおいてWTー1対応遺伝子を単離し、ES細胞系の同遺伝子に相同組換えで欠損を導入した。
