ラット胸腺腫発生遺伝子Tsrー1の作用機序に関する研究

研究概要	1.Tsrー1遺伝子導入コンジェニックラットの樹立:BUF/Mnaラットの胸腺腫感受性遺伝子Tsrー1を胸腺腫嫌発系ACI/NMsラットに導入するため、胸腺腫発生を指標として、退交配を続け第9代に達した。 2.Tsrー1遺伝子の発現の場の検索:老齢のBUF/Mnaラットに致死量の放射線を照射し、異種であるBALB/cヌ-ドマウスの骨髄細胞を4X10^7個静注した。4週後の屠殺時に胸腺腫の再構築が認められ、胸腺腫内のリンパ球は95%以上がマウスThyー1.2を発現していた。従って、胸腺腫の本体は上皮細胞であることが実証された。 3.胸腺腫上皮細胞の分子生物学的検索:(1)ラット正常胸腺、胸腺腫及び胸腺癌由来の合計7株の上皮細胞株について、Northern blottingにて種々の癌関連遺伝子および癌抑制遺伝子の発現の有無を検索した。cーmyc、p53はすべての細胞株で同レベルの発現を認めた。EGFーRは正常胸腺及び胸腺腫由来の細胞株で発現がみられたものの、胸腺癌由来細胞株では低レベルであった。TGFーβについては逆に胸腺癌由来細胞株で発現が増強していた。(2)正常胸腺および胸腺腫の初代培養において、各種のサイトカインのmRNAの発現を検索したところ、胸腺腫特異的にGーCSFの高い発現が見られた。(3)Tsrー1とrnu遺伝子の連関が示唆されたので、両遺伝子に関するcongenic rat、Eviー2及びMpo遺伝子のプロ-ブを使用して、Tsrー1のクロ-ニングに着手した。 4.胸腺腫内リンパ球の生理的細胞死の障害:10%FCS添加RPMI培養液中でWistar系ラット胸腺リンパ球を培養すると24時間後には約30〜40%しか生存していないが、BUF/Mna系ラット胸腺腫内リンパ球は55〜65%の細胞が生存する。この生細胞数の高度残存は、BUF/Mna系ラット胸腺腫内リンパ球の生理的細胞死機構が障害されていることを示すものと考えられる。また、老令BUF/Mna系ラットにおいて、抗TCRαβ抗体あるいは抗CD3抗体と抗マウス免疫グロブリン抗体により細胞表面上のTCR複合体を架橋したところ、細胞内Ca濃度の上昇が認められた。従って胸腺腫内リンパ球のTCRは刺激伝達能を保持していることが明らかになった。
