研究概要

本年度は計画の2年目にあたるが、計画時に解明したいと考えていた骨髄や胸腺ストロ-マ細胞とリンパ系幹細胞の相互作用に関する大筋についてはほぼ理解できたのではないかと考えている。
骨髄中でストロ-マ細胞が発現してB細胞の増殖を調節している分子については、ILー7,cーkitのリガンドであるSLFが固定された。しかし、西川らの研究によってSLFとILー7のみではストロ-マ細胞そのものを用いた培養に比較してたかだか1/10程度の増殖しか見られない事が明らかにされており他の分子の関与が示唆された。実際、菊谷らはストロ-マ細胞上のB細胞株の増殖を抑制するモノクロ-ナル抗体を樹立しており、この分子の解明が待たれる。一方B細胞分化のシグナルについては、仲野らの様にストロ-マ細胞由来とする実験結果と、西川らのB細胞自律的であるとする考えがあり、今後の課題であろう。
胸腺内ストロ-マ細胞の役割についての解明もかなり進んだ。八木田、緒方、宮崎らの研究の結果、MHC分子、接着分子を介するT細胞増殖調節が可能な事や、ILー2、ILー7などのサイトカインの役割が明らかにされた。しかし、垣生らのSCIDマウス胸腺に関する研究は、胸腺内での細胞増殖シグナルの最も重要な部分がT細胞レセプタ-を介している事を示唆している。この点の確認については現在T細胞レセプタ-トランスジェニックマウスを用いて検討中である。
以上の様にリンパ系幹細胞の増殖支持に関わる分子の大半については明らかになったが、造血幹細胞からリンパ系へのコミットメントの問題や、後期の分化に関するシグナルの実体、さらには細胞表面レセプタ-以降のシグナル伝達経路など、新たな問題が出ており、次年度はこの方向の研究に必要な実験システムを地道に開発する必要があろうと考えている。
