研究概要

伊東はDMAB前立腺癌発生モデルを用いて、テストステロンが前立腺癌の強力な促進作用を有することを確認するとともに、腹葉前立腺以外で異形成から初期癌そして浸潤癌へ至るプロモ-ションからプログレッションの過程にテストステロンが極めて重要な働きをしていること、異型過形成が前立腺癌の前癌病変であることを明らかにした。立松はこれまで困難とされていたマウスの腺胃癌の作成にMNUを用いることにより成功し、ヒト胃癌に類似した分化型腺癌の転移のモデルとしてこれが応用可能であることを示した。石川はヒト子宮頚部進行癌で癌抑制遺伝子の欠失を高頻度に認め、進行癌と癌抑制遺伝子の欠失との関連性を示した。江角はヘテロサイクックアミンによるZymbal腺腫瘍発生でMeIQxとIQはともにHーrasの活性化が重要な役割をはたしていることを明らかにし、さらに両者ではその活性化で塩基配列に対する特異性に差があることを示した。伊藤は甲状腺濾胞上皮細胞に対して、エストロジェンだけでなくてテストステロンでもその受容体を通してプロモ-ション作用を示し、下垂体TSHは別の膜受容体を介して作用し、甲状腺発癌のプロモ-ション作用を強くすることを示した。岡田は癌細胞の悪性化の進展を促進する要因として、宿主反応細胞、さらにこの反応細胞から放出される内因性の活性酸素が関与することを明らかにした。
本年度の研究で潜在韓の顕在化に関与する要因として、エストロジェンやテストステロンの受容体変異の増大、癌細胞などが放出するprostaglandin E_2などアラキドン酸代謝に関与する要因や活性酸素の関与、また、癌遺伝子の活性化や癌抑制遺伝子の欠失などをヒトの子宮頚癌やヘテロサイクリックアミンにより誘導される腫瘍について明らかにした。これらの変化は多段階発癌に伴う諸変化で潜在癌の顕在化に関する新しい知見が得られたものと考えられる。
