細胞増殖制御因子および耐性因子に作用する制癌剤の探索とその応用

研究概要

癌化学療法を発展させるために新しい作用および構造を有する制癌剤の発見を目的に研究を実施し、本年度は以下の研究成果を挙げた。
坑生物質azatyrocineは癌遺伝子によって癌化した細胞の増殖を特異的に抑制し、revertant細胞を誘導する。Azatyrosineによって誘導されたrevertant細胞に特異的に発現している遺伝子を解析した結果、抑制遺伝子rrgの他、rho B、collagen、IAP関連遺伝子他が見出された。この中でrrgは正常細胞に多量に発現していることなどから、細胞の正常化を維持する為に重要な遺伝子であることが示唆された。CーHaーrasを組込んだtransgenicマウスを用いてazatyrosineの発癌予防実験を行った結果、azatyrocineは前胃へのパピロ-マ形成を著明に抑制した。PIP_2から情報伝達物質IP_3およびDGの生成を触媒するphospholipas Cの特異的阻害物質を微生物代謝産物中に探索し、Q12713物質を発見した。本物質は強い細胞毒性を示し、固型癌にも有効な物質として得られた既知物質であるが、in situにおける作用機作に興味がもたれる。T細胞活性物質として放線菌生産物より発見されたconageninは細胞障害性T細胞ならびにNK細胞など抗腫瘍エフェクタ-の生成を増強し同系腫瘍に制癌効果を示した。ILー3産生の増強を示すことから骨髄機能に対する作用を検討したところ、抑制された末梢血中血小板数の回復を示した。固定癌に有効な物質を得ることを目的に血管新生阻害物質を探索し、ビタミンD誘導体にその活性を見出し、DMBA誘発乳癌に対する制癌効果を調べた結果、著明な増殖抑制効果がみられた。脂肪酸の制癌作用に着目し、ソルビト-ルにオレイン酸を酵素学的手法によって結合したソルビト-ル・モノオレ-トに制癌ならびに著明な転移抑制効果を認め、TNFとの併用効果もみられた。以上の各物質につきさらに詳細な作用ならびにその機作を追究するとともに新物質の探索を続けている。
