成人T白血病細胞株に認められたアルギニノコハク酸合成酵素遺伝子発現の異常亢進;ーその白血病診断マ-カ-としての意義ー

研究概要	アルギニノコハク酸シンテタ-ゼ(ASS)は尿素サイクルを構成する5つの酵素のうちの1つで、尿素サイクルの律速段階を仲介している。このASSはおもに肝臓において高い活性を示し、組織非特異的に広く存在する。本研究では、このASS遺伝子発現が白血病の診断マ-カ-として有用であるかどうかを検討した。その根拠はヒトの正常リンパ球では殆どASS遺伝子の発現が認められないが、一部の白血病患者できわめて高いASS遺伝子の発現が検出されたことによる。方法は半定量的なASS遺伝子のRTーPCR法が用いられた。24例の種々の白血病患者及び15例の健常人の末梢血リンパ球について検討した。内訳は、急性リンパ性白血病(ALL)3例、急性骨髄性白血病(AML)4例、慢性骨髄性白血病(CML)の急性転化例2例、成人T白血病(ATL)7例、慢性リンパ性白血病(BーCLL)2例、CML3例、真性赤血球増加症1例である。その結果、健常人に比較し、AML,ALL及び急性転化したCMLで10〜30倍のASS遺伝子の発現亢進が認められた。以上の結果から、1)ASS遺伝子発現が単に細胞のトランスフォ-メ-ションや細胞増殖のメカニズムによるのではなく、ALLやAMLの白血病化の分子機序と関連していること、および2)「ASS遺伝子マ-カ-」はCMLからの急性転化を予知する方法として有用である可能性が示された。分子生物学的予知法としては初めてのものである。
