研究概要

ヒト肝細胞増殖因子(human hepatocyte growth factor;hHGF)は、ラットなどの実験動物のそれとは血中での存在様式が異なり、またラットやマウスのhHGFは、我々が調製したhHGFに対する抗体とも交差反応性を示さない。そこで本研究は、hHGFに対する抗体と交差反応性を示すサル及びヒトの組織を用いて行った。なお、ヒトの組織は本学医学部附属病院での各種切除手術及び病理解剖等で得られた試料を用いた。その結果、1)正常のヒト及びサルの肝臓ではほとんどhHGFのmRNAが検出されないが、劇症肝炎患者及び四塩化炭素投与後のサル肝臓ではhHGFのmRNAが高濃度に検出される。2)正常ヒト及びサルの脾臓を、hHGFのcDNAをプロ-ブとしてin situハイブリダイゼ-ションによる染色を行ったところ、いずれもpositiveな細胞が見られたが、銀粒子にマスクされ細胞を同定することが出来なかった。3)正常及び四塩化炭素投与サルの脾臓をhHGFに対する抗体を用いて免疫組織的に染色したところ、染色性のあるものは血球系の細胞と判定された。
以上のことから、肝疾患患者血漿中に出現するhHGFは、肝障害のシグナルによって脾臓から肝臓に動員された血球系の細胞が合成分泌しているものと推定された。現在は劇症肝炎患者から得た試料により、以上の推定を最終的に検討しているところである。上記の研究と平行して、研究代表者らはBirchmeier教授(ドイツ、エッセン大学癌研究所)との共同研究により、従来癌細胞の転移と関係があるとされてきたscatter factorがhHGFと同一蛋白であることを明らかにすると共に、hHGFが悪性度の高い肝癌細胞のDNA合成を抑制する作用を見出した。
