環境変異原と重金属化合物との相互作用によるヒト遺伝子損傷と複合汚染における意義

研究概要	本研究の目的はAmesテスト陰性の発がん物質と金属との相互作用によりヒト偉伝子が損傷される機構を解明することである。本年度は種々のニッケル化合物、およびトリプトファン代謝物の一つである3-ヒドロキシルアントラニル酸(3-HAA)を選び、DNA分子レベルおよび細胞レベルでヒト偉伝子損傷を検討した。細胞レベルでのDNA損傷はパルスフィールドゲル電気泳動を行ない測定した。ニッケル化合物(1)ヒト培養細胞(Hela、Raji細胞)を硫化ニッケル(NIS、Ni_3S_2)で処理すると二本鎖DNA切断が認められた。アルカリ処理でDNA切断が増加することから、一本鎖DNA切断も推定された。(2)カタラーゼ阻害剤、GSH合成阻害剤やGSH消去剤で、NiSによる二本鎖DNA切断が増加したことから、過酸化水素の関与が示唆された。(3)Ni_3S_2で処理したHeLa細胞では8-OH-dGが有意に増加した。このことは我々がin vitroで明かにした酸素ラジカルによるDNA損傷が細胞内でも起こる可能性を示している。3-HAA(1)3-HAAはCu(II)の存在下でDNA切断をもたらした。カタラーゼとバソキュプロインの添加によりDNA損傷が抑制されることから、H_2O_2とCu(I)の関与が示された。OHラジカル捕捉剤による抑制は少くな、HOラジカルがDNA損傷に果たす役割は小さいと考えられる。(2)3-HAAにMn(II)を加えた後Cu(II)を加えたDNAの方がMn(II)のない場合に比べてDNA損傷が大きかった。(3)3-HAAはCu(II)の存在下でcalf thymus DNA中の8-OHdGを増加させた。3-HAAとCu(II)から生じたH_2O_2とCu(I)から活性酸素が生成しDNAを損傷することが示唆された。Mn(II)はH_2O_2の生成を促進すると考えられる。
