研究概要

生体において抗体産出細胞等の特定の細胞が活性化される機構を分子レベルで理解し、それに基づいて新しい生体賦活化材料の作成を推進すること、並びに生体賦活因子の生体ほの効率的な送達法を確立することを目的として、総合的な検討を行い、以下に示す成果を得た。(1)薬物のイオン化率と皮膚透過の関係ならびに透過に対するテルペンの影響について検討した結果、薬物の皮膚透過性は、ほぼpH分配仮説に従うこと、またlーメント-ルはイオン型薬物の皮膚透過に対する促進作用に優れることを見い出した。これに対し,dーリモネンは分子型薬物の透過を強く促進し、この差はテルペン分子の疎水性に依存することを明らかにした(永井)。(2)動物細胞であるWTAラットの白血病細胞およびフィッシャ-ラット系の胸腺腫細胞とPEG脂質含有リポソ-ムとの相互作用を、蛍光法および蛍光顕微鏡観察により検討した結果、融合の機構でリポソ-ム多糖が取り込まれている可能性が示唆された。また多糖でリポソ-ムを被覆することにより、食胞効率の制御が可能となった(砂本)。(3)免疫賦活活性を持つ多糖と、免疫アジュバントであるムラミルジペプチドとのコンジュゲ-トによる高分子ドラッグを合成し、活性をin vitroで評価した結果、コンジュゲ-ト化によって抗腫瘍活性の向上が認められた。in vivoでの腫瘍移植マウスの延命効果についても良好な成績が得られている(大内)。(4)温度応答性ゲルをスイッチとして利用することにより、薬の放出を完全にONーOFF制御できることを見い出し、またこのプロセスの理論的な解明に成功した(岡野)。(5)光駆動性クロライドポンプ機能を持つハロロドプシン(hR)をLB膜中に固定化させる方法について検討した結果、膜脂質単分子膜にhRを融合させる方法およびhRを直接LB膜化させる方法のいずれにおいても製膜が可能であることを明らかにした(久保)。
