シナプス小胞と原形質膜との融合過程における低分子量GTP結合蛋白質の役割

研究概要

昨年度までに私共は低分子量GTP結合蛋白質(G蛋白質)の一種であるsmg p25Aを見出し、smg p25Aが神経組織のシナプスに大量に存在していることを明らかにしている。さらに、smg p25AのGDP/GTP交換反応を抑制する蛋白質(GDI)を発見し、このsmg p25A GDIはsmg p25Aのシナプスにおける膜画分と可溶性画分のトランスロケ-ションを制御することも明らかにしている。ras p21において、そのC末端側の翻訳後修飾がras p21の機能発現に重要であることが報告されているので、本年度は、まずsmg p25AのC末端側の翻訳後修飾を解析した。smg p25AのC末端側のアミノ酸配列はCysーAlaーCysであるが、この両方のCys残基はゲラニルゲラニル化され、C末端のCys残基はメチル化されていた。このsmg p25Aの翻訳後修飾の機能を明らかにするために、ウシ大脳膜画分から精製した翻訳後修飾のあるsmg p25Aと、大腸菌から精製した未修飾のsmg p25Aを用いて、それらの性状を解析した。その結果、smg p25Aの翻訳後修飾はsmg p25Aの細胞膜への結合やsmg p25A GDIの作用に必須であり、翻訳後修飾の中でも特にゲラニルゲラニル化が重要であることが明らかになった。したがって、シナプス小胞と原形質膜との融合過程にsmg p25Aが作用する際にもsmg p25Aの翻訳後修飾が重要な役割を果たす可能性が高いと考えられた。さらに、私共はウシ大脳の可溶性画分からsmg p25Aに対するゲラニルゲラニル基転移酵素(GGT)を部分精製した。私共はすでにsmg p21/rhoA p21に対するGGTを精製していたが、smg p25A GGTはsmg p21/rhoA p21をゲラニルゲラニル化せず、また、smg p21/rhoA p21 GGTはsmg p25Aをゲラニルゲラニル化しなかった。したがって、smg p25A GGTとsmg p21/rhoA p21 GGTは異なる酵素であることが明らかとなった。以上の結果を得たことにより、本年度の研究計画はほぼ達成できたと考えている。
