金属プロテア-ゼの構造と機能

研究概要	クサリヘビRussell's viper毒より、血液凝固X因子及びIX因子を特異的に限定分解し活性型に変換する酵素のRVVーXを精製し、その全アミノ酸配列を決定した。RVVーXは二本鎖より成り、重鎖(59kDa)と軽鎖(LC1=18kDa/LC2=21kDa)がSーS結合していた。LC1は123残基のアミノ酸から成り、24位のアスパラキンには糖鎖を含んでいる。NBRFデ-タベ-スを用い相同性検索を行なった結果、意外なことに、LC1は肝臓のアシアロ糖蛋白受容体やセレクチンのいわゆるC型レクチンと相同性のあることが見い出された。LC1は半シスチンを7残基持つが、SーS結合が既に決定されているフジツボレクチンとの相同性よりCys4ーCys15、Cys32ーCys121、Cys98ーCys113の3組がSーS結合し、また、LC1に特異的なCys77は重鎖とのSーS結合に関与すると推定した。一方、重鎖はハブ毒の高分量出血因子HR1B(415残基)と酷似していた。すなわち、重鎖はN末端半分の領域に金属プロテア-ゼドメインを含み、亜鉛を配位するリガンドのHis145、His149及び活性残基の一部と考えられているGlu146も保存されていた。一方、C末側には強力な血小板凝集阻止因子であるdisintegrin様の構造ドメインを含んだ。したがって、RVVーXは、軽鎖にC型レクチン様ドメインを、重鎖に金属プロテア-ゼとdisintegrin様ドメインを含む新しいプロテア-ゼであることが明らかとなった。今後は蛇毒の金属プロテア-ゼの高次構造(目下、阪大タンパク研との共同研究でこの種のプロテア-ゼの結晶回析を進めている)をもとに、Zn^<2+>の配位した触媒基の同定及び反応機構、特異性に関連した構造要素などを明らかにしたい。
