温度感受性変異レトロウイルスを用いた造血幹細胞株の樹立と分子生物学解析

研究概要

我々が分離したAbelsonマウス白血病ウイルスの温度感受性変異株(tsmutant)でトランスフォ-ムした未熟B細胞株では、許容温度(35℃)では、増殖するのみであるが、非許容温度(39℃)に培養温度を上げると、細胞が、transformedの状態から、untransformedの状態に変わり、このため分化が一斉にはじまり、まず1g遺伝子の再構成がおこり、次に、T細胞レセプタ-遺伝子の再構成も起こる。さらに細胞表面には、Thyー1抗原が一過性に出現したり、RA3ー6B2モノクロ-ナル抗体で検出されるB220抗原も、cyclicに発現した。このようなデ-タから、血球系細胞の分化研究に対する、Abelsonウイルスのts mutantの有用性が確立された。そこで、このts mutantを用いて、血球系のより未熟な細胞をトランスフォ-ムさせ、stem cell lineを樹立し、stem cellの分化を、培養温度でコントロ-ルし、その分化機構を解明しようとするのが本研究である。現時点での研究状況は次のようである。1.マウスの骨髄細胞からDexture cultureを作製した。一方、ts vーabl geneを挿入したpZip・neoーtsーvーabl retroviral vectorやLTRをMPSVにかえたvectorを作製した。これらのretroviral vectorを、Dexture cultureに感染させ、トランスフォ-ムし株細胞の樹立を試みているが、今のところ、stem cell lineの樹立には成功していない。2.上記のretroviral vectorを正常マウスの骨髄細胞に感染させ、致死的に放射線照射したマウスに骨髄移植を行ない、白血病化を試み、これから、stem cell line樹立を目ざしている。
