研究概要

(1)洞結節細胞のペ-スメ-カ機能をシミュレ-トする際に重要となる背影電流に占めるNaーKポンプ電流の寄与を検討したところ、このポンプ電流は緩徐脱分極相で10〜26PAに達することが判明した(入沢)。(2)作業心筋の興奮伝導を規定するNa電流モデルを単離心室筋細胞の膜活動電位立ち上がり相と生理的な条件下(37℃、正常Na勾配)での膜電位固定によるNa電流記録に基づいて構成し、これをギャップ結合(100〜1000ns)を介して格子状にあるいは六角状に配列して興奮伝導をシミュレ-トしたところ、後者の配列の方がモルモット乳頭筋標本から得た二次元の興奮伝播様式をより良く表現することが判明した(外山)。(3)Di Frances co & NobleのPurkinje線維の膜活動電位モデルを基本として、心室筋の再分極過程モデルを実験的に得られたI_K、I_<TO>、I_<KCa>、I_<cl>から構成したところ、細胞内Ca濃度による修飾が重要であることが判明した(沢登)。(4)簡略化したイオン電流モデルまたは心筋の膜活動電位波形を二次元またはヒトの心臓を模した構築を持つ素子に抗して、不整脈の発生機構をシミュレ-トした。不整脈の発生には時空間的な興奮伝導性の不均一化が重要であり、特に心室細動発生はPurkinje線維の心拍数に依存した不応期の変化をイヌの実験デ-タに基づいて心室筋のそれの数倍に認定する事が致死的不整脈である心室細動発生の必要条件となる(春見、武者、安井、鈴木)。(5)イヌ冠動脈に直径25μm以上のマイクロフフェアを注入すると組織標本上でパッチ状の梗塞がみられ、心室遅延電位が出現した(井上)。
