ADF/Thioredoxinによる発ガン遺伝子発現の制御

研究概要

本年度我々は細胞の活性化、癌化における酸化還元調節因子ADF/Thioredoxin(TRX)の細胞内活性を以下のような点について検討した。
1)ADF/TRXが転写制御因子の活性化機能を持つことを、1Lー2Rα鎖の転写調節をモデルとしてin vitro,in vivoの系を用いて証明した。
2)ADF/TRX reductaseの特異的阻害剤である13ーcis retinoic acid(RA)及びジチオ-ル基特異的阻害剤であるNーetylmaleimide(NEM)を用いて細胞内ADFの役割について検討を行ない、これらのADF阻害剤はADF高産生細胞株であるHTLVー1陽性細胞の増殖を抑制することを証明した。
3)ATLを代表とするある種のウイルス発ガンおいてADFが高発現することから、ウイルスによるADF遺伝子発現制御の解析が必要となることなる。そこでADFの発現制御領域の解析を目的としてgenomic ADFのクロ-ニングにより複数のクロ-ンを得た。
4)ヒトADFcDNA及びβーactinのpromotorとterminatorを用いてヒトADF発現ベクタ-を構築し、B6マウスを用いてtransgenic mouseを作製した。
