研究概要	サイトカイン系におけるシグナル伝達機構の解明を目的とし、インタ-ロイキン2(ILー2)系をモデルシステムとして用い、ILー2受容体β鎖を介する細胞内情報伝達機構の解析を行った。昨年度にはβ鎖の下流に位置しβ鎖と共役するシグナル伝達分子として、リンパ球特異的に発現しているsrcファミリ-チロシンキナ-ゼp56^<lck>を同定し、加えてこれら分子間の相互作用に必須な領域を両分子において同定した。本年度は更に、p56^<lck>とβ鎖の会合がILー2による細胞内タンパクチロシンリン酸化亢進に必須であることを明らかにした。しかしながら、β鎖を発現させた細胞に活性型p56^<lcl>を導入してもILー2依存性を失わないことからILー2による増殖性シグンル伝達にはチロシンキナ-ゼ経路以外のものも重要であることが考えられる。ILー2によって核内プロトオンコジ-ンcーfos,cーjun及びcーmycのmRNA発現が効率良く誘導されるが、“acidic region"を欠失させたβ鎖はcーfos,cーjunのmRNA発現を誘導することが出来なかった。一方、“serineーrich region"を欠失させたβ鎖についてはいずれかのmRNAの誘導もほとんど検出されなかった。このことからβ鎖各細胞内領域と、これら遺伝子の誘導には機能的連関があることが考えられる。サイトカイン遺伝子の発現機構の解明を目的とし、インタ-フェロン系においてその解析を行った。本年度は転写調節因子IRFー1がIFNーβ遺伝子及びINFー誘導遺伝子の発現に果す役割を詳細に検討する為、IRFー1cDNAのアンチセンスまたはセンスRNAを強制発現する細胞株を樹立した。センスRNAを発現する細胞株では、コントロ-ル細胞に比べ、polt(rI):(rc)刺激及びウイルス感染によるIFNーβの誘導ならびにIFNー誘導遺伝子(MHCクラスI分子)の発現は顕著に増加した。一方、アンチセンスRNAを発現する細胞株ではこれらの遺伝子の誘導が抑制された。このことから、IRFー1が実際に細胞内でIFNーβおよびMHCクラスI遺伝子の発現においてポジティブに作用していることが明かとなった。
