研究概要

マウスIL‐5レセプタ-α鎖(IL‐5結合分子)cDNAをプロ-ブに、プラ-クハイブリダイゼイション法により、ヒト好酸球cDNAライブラリ-よりヒトIL‐5レセプタ-α鎖のcDNAを単離しその全構造の決定をした。ヒトIL‐5レセプタ-α鎖も膜貫通型のものと可溶性型の2種類あり、そのいづれもメッセ-ジが好酸球で発現していることが確かめられた。リコンビナント ヒトIL‐5レセプタ-α鎖はマウスのα鎖とよく類似しており(アミノ酸レベルで約60%の類似性あり)、細胞質外トメイン、膜貫通部分、および細胞質内ドメインより構成されていた。細胞質外ドメインには4コのシスティン残基モチ-フWS×WSモチ-フがそれぞれ保存されており、サイトカインレセプタ-に共通の構造をしていた。細胞質内ドメインは比較的短かく、チロシンキナ-ゼドメインやキナ-ゼのカタリティックドメインは存在せず、α鎖単独ではIL‐5シグナルが伝わらないことを予測させた。このcDNAをCOS細胞に発現させると比較的高い親和性でIL‐5と結合する分子が発現した。又IL‐3依存性FDC‐P1にヒトIL‐5レセプタ-α鎖を発現させるとヒトIL‐5に応答性を獲得しDNA合成を示した。このことよりヒトIL‐5レセプタ-α鎖もマウスIL‐5セレプタ-β鎖(Alc2B)と会合し、IL‐5に応答性を付与させうると考えられ、ヒトα鎖もヒトβ鎖と会合し機能的IL‐5レセプタ-を構築していると予測された。リコンビナントIL‐5α鎖の分子量は約60〓であった。今後はヒトIL‐5レセプタ-β鎖の同定とそのシグナル伝達の解析が必須と思われる。
