痛覚関連物質mRNAの動的解析

研究概要

脊髄への入力と出力を切断したときに生じる脊髄の細胞組織構築と、それに伴う痛覚関連神経伝達物質の変化を、ペプチドと蛋白については免疫組織化学法で、またmRNAについてはin situハイブリダイゼ-ション法を用いて解析した。成熟ラットの脊髄前根と後根を切断すると、伝達物質(CGRP,サブスタンスP,NPY,ガラニン,エンケファリン,セロトニン)はそれぞれに特有のパタ-ンを示しながら変化していった。脊髄根を切断することで、脊髄灰白質および白質の組織構築に変化が認められた。とくに微小管関連蛋白ー2(MAPー2)の分布は灰白質に限局して存在するため、MAPー2免疫反応陽性の灰白質の面積の変化が画像解析装置を用いて定量的に示された。また、胎生ラット脊髄を成熟ラットの前眼房内に移植し、伝達物質の分布の変化を観察したが、in vivoの発育と同様のパタ-ンを示すものと、in vivoとは異なる様相を呈するものに分かれた。以上から、脊髄内、とくに後角に存在する神経伝達系には明らかに外界、内界の環境変化にともなうheterogeneityが存在することが示された。このことによって、痛覚機構に関与するそれぞれの伝達物質は、刺激に対して単純な反応様相を呈するのではないことを表わしていると結論づけられた。
