シロイヌナズナを用いた光合成遺伝子発現に介在するシグナル伝達機構の解析

研究概要

突然変異体の作製・解析および変異相補という方法論は,シグナル伝達機構の解明において極めて強力な研究手段である.この種の方法論の適用が可能であるモデル植物シロイヌナズナ(Arabidopsis thaliana)を実験に共した.
<1.選抜のための遺伝子の植物体への導入>___ー
光合成遺伝子としてリブロ-ス-1.5ー二リン酸カルボキシラ-ゼ/オキシゲナ-ゼ(RubisCO)のSサブユニット遺伝子(rbcS)およびクロロフィルa/b結合タンパク質遺伝子(cab)に着目した.校正活性を有するDNAポリメラ-ゼを用いたDNAポリメラ-ゼ増幅法(PCR)により,シロイヌナズナrbcSー3Bおよびcab1およびcab1のプロモ-タ-とそれらの上流を含むそれぞれ約1,500ヌクレオチドからなる領域を作製した.これらプロモ-タ-の下流にreporter遺伝子としてβーglucuronidase(GUS)あるいはchloramphenicol acetyltransferase(CAT)遺伝子を繋ぎ,シロイヌナズナ(ColumbiaおよびNorway)への導入を試みた.また,別途上記PCR法により増幅したアルコ-ル脱水素酵素遺伝子(adh)をrbcSー3Bプロモ-タ-の下流に繋ぎ,シロイヌナズナ(Bensheim,adh^-)への導入を試みた.
<2.突然変異体の作製と選抜>___ー
上記形質転換植物体から応られる種子を変異原ethylmethanesulfonate(EMS)により処理し,得られるM_2個体のうちホモ接合よって光合成能が低下した突然変異体を選抜する.光合成突然変異体が長波長紫外光の照射により赤色螢光を発することを確認した.また,adh遺伝子発現が低下している植物体は,allyl alcoholを植物毒性アルデヒドacroleinに変換できないためallyl alcohol耐性であった.
