蛋白質の選別輸送における低分子量GTP結合蛋白質の機能と作用機構

研究概要

昨年度までに私共は、私共が発見した低分子量GTP結合蛋白質(G蛋白質)smg p25Aがシグナル依存性の分泌細胞に存在することを見出し、さらにsmg p25AのGDP/GTP交換反応を抑制する蛋白質(GDI)を発見し、このsmg p25A GDIがsmg p25Aの膜画分と可溶性画分のトランスロケ-ションを制御することを明らかにしている。現在、蛋白質の選別輸送に低分子量G蛋白質が関与していることは確定しているが、低分子量G蛋白質の標的蛋白質は明らかになっていない。そこで本年度は、まずsmg p25Aの標的蛋白質を同定することを試みた。その結果、GTP結合smg p25Aと結合する蛋白質をウシ大脳膜画分から部分精製することに成功した。本蛋白質の分子量は約86,000であり、GDP結合型smg p25Aとの親和性はGTP結合型に比較すると低かった。また、本蛋白質は他の低分子量G蛋白質(Kiーras p21,rho A p21,smg p21,rab 11 p24)と結合しなかった。さらに,本蛋白質にはsmg p25AのGDP/GTP交換反応を制御する活性やGTPase活性を促進する活性は認められなかった。したがって、本蛋白質はsmg p25Aの活性制御蛋白質ではなく、GTP結合型smg p25Aと特異的に結合するsmg p25Aの標的蛋白質であると考えられた。一方、ras p21のC末端側の翻訳後修飾がras p21の機能発現に重要であることが報告されているので、次に、smg p25AのC末端側の翻訳後修飾を解析した。その結果、smg p25AのC末端側のアミノ酸配列はCysーAlaーCysであり、この両者のCys残基がゲラニルゲラニル化され、さらに、C末端のCys残基がメチル化されていることを明らかにした。このsmg p25Aの翻訳後修飾はsmg p25Aの細胞膜への結合やsmg p25GDIの作用に必須であった。したがって、smg p25Aの翻訳後修飾はsmg p25Aが標的蛋白質に作用する際にも重要である可能性が高い。来年度は、smg p25Aの標的蛋白質を均一蛋白質に精製し、蛋白質の選別輸送におけるsmg p25Aとその標的蛋白質やsmg p25GDIの役割を明らかにする予定である。以上の結果を得たことにより、本年度の研究計画はほぼ達成できたと考えている。
