研究概要

本年度の成果は以下の通りである。
カルモジュリン遺伝子は小核から大核に分化する過程で5'上流域の欠失が起こることが判明した。また、ゾウリムシの大核分化時にカルシウムイオンが密接に関与していることが示唆された。さらに、ミドリゾウリムシでは、未熟期の長さ、接合活性リズムなど接合型の継時的発現に関する変異体が得られた。一方、短寿命変異体の増殖速度を回復させるParGF(ゾウリムシ成長因子)活性をもつ物質として、哺乳類由来の表皮成長因子、トラスフォーミング成長因子、テトラヒメナ由来のcrude sampleが新たに発見された。また、MPF活の探索の過程でゾウリムシとテトラヒメナの細胞抽出液中にcdc2ホモログが存在することも判明した。
接合型(E,O)の分化に関し、シンジェン(同胞種)間での大核クロマチン移植を行なったところ、donorの優性形質(E)が現れずに劣性のrecipient(O)が自身の優性形質(E)を示し、接合型決定遺伝子のシンジェン特異性が転換することが示唆された。ゾウリムシの口部装置側繊毛特異的モノクローナル抗体が得られ、繊毛は細胞の場所によって物質最に分化していることが初めて明らかになった。一方、テトラヒメナの染色体Gバンド作成がほぼ完了した。また、接合過程の解析に役立つ様々な染色体変異株、および、in vivo染色体切断法による欠失株の系統的作成法などを開発した。
大核DNAの制限酵素断片による形質転換法を用いて、いくつかの遺伝子を含む断片の紋り込みが進んだ。高率な転換には、recipientの細胞周期が重要であることもわかった。また、形質転換用ベクターの候補であるミトコンドリア内プラスミド様DNAはヘアピン状構造を持つこと、野外株ごとに多様化していること等がわかった。
