研究概要	筋形成について、筋の前駆体である中胚葉の誘導から、細胞分化、収縮構造をもつ機能細胞の形成に至る各段階の分子機構を検討した。 1)浅島はアクチビンの中胚葉誘導作用を既に発見したが(1990)、今年度、中胚葉(筋)誘導に適切なアクチビンの濃度域の決定、両性類の未受精卵におけるアクチビンの存在、フォリスタチンのアクチビンへの結合による機能抑制および初期胚にフォリスタチンの存在を見いだした。 2)筋細胞系列の決定と分化について、原索動物ホヤをモデルとして筋細胞決定・分化因子を探索し、PCR法により脊椎動物の筋分化因子であるMyoDに似た構造をもつ遺伝子の断片の単離に成功した(佐藤)。遠藤はマウス筋細胞株(C2細胞)にSV40Tを発現させて脱分化を誘起する独自の系を用い、筋細胞の脱分化時にc‐jun、cdc2の発現誘導とRBの発現抑制がおこることが見いだした。また転写因子AP‐1(c‐Jun/c‐Fos複合体)の発現増強がMyoDの転写抑制を引き起こすことを示した。 3)ニワトリ胚の体筋内筋芽細胞を蛍光標識して、筋芽細胞の肢芽筋形成領域への移動過程、また移動を導く要因を明らかにした。 4)筋蛋白質遺伝子とその転写制御について、鍋島はミオシン軽鎖遺伝子の領域MLCboxへの制御因子の結合による遺伝子の転写促進、またMyoDとmyogeninが異なる因子との相互作用を介しEーboxに作用して転写を誘導することを明らかにした。嶋田はニワトリ骨格筋で心筋トロポニンC発現が転写されるが、翻訳されない事実を見いだした。佐藤はホヤアクチン遺伝子の染色体上の局在、ミオシンH鎖の構造を決めた。 5)筋蛋白質の集合について、眞埼は筋発生初期に特異的な新たなαアクチニンを見いだし、細胞内局在を解明、大日方はアクチン集合制御因子として、胚筋のプロフィリンのcDNAの構造を決定、また筋型、非筋型コフィリンを識別、またコフィリンの機能ドメインを明らかにした。
