研究概要

シナプスにおける化学的情報伝達に関して、以下のような研究成果をあげた。
1.神経伝達物質受容体の機能と分子構造 三品はクロ-ニングしたマウス脳の非NMDA型グルタミン酸受容体cDNAに点変異を与え、受容体のCa^<2+>透過性が劇的に変化することを明らかにした。小澤は海馬ニュ-ロンでAMPAとカイニン酸が同一のグルタミン酸受容体を活性化することを明らかにした。小野寺はラット脊髄ニュ-ロン上のグルタミン酸受容体の分布状況を明らかにした。赤池はマイネルトニュ-ロンでGABA_B受容体を介してGABA遊離が抑制されることを示した。高橋はグリシン受容体には成熟型と幼若型が存在し、両者でチャネルの開口時間に100倍以上の差があることを示した。東は前頭前野ニュ-ロンの興奮性および抑制性シナプス機序を調べ、それぞれグルタミン酸とGABAが伝達物質であることを明らかにした。
2.細胞内情報伝達系による受容体活動の制御 久場は共焦点レ-ザ-顕微鏡を駆使してニュ-ロン内のCa^<2+>濃度の空間的、時間的変動の記録法を確立した。赤須はカルモジュリン依存性プロテインキナ-ゼの活性化に伴うリン酸化によるMチャネルの制御機構を明らかにした。
3.伝達物質放出の制御機構 立花と八尾はそれぞれキンギョ網膜双極細胞、鶏胚毛様体神経節のシナプス前終末部の伝達物質放出に直接関係する膜電位依存性Ca^<2+>チャネルの性質を詳細に解明した。
4.シナプスの可塑性発現機構 杉山はGタンパク質共役型グルタミン酸受容体が海馬の長期増強発現に関与することを示した。工藤は海馬スライスで長期増強の発現とCa^<2+>濃度上昇との関連を調べ、長期増強の発現機構を解明するための基礎デ-タを得た。平野は小脳培養系を活用して、長期抑圧の発現機構を明らかにした。
