研究概要

シナプスの化学情報伝達機構の研究で特に次の二点において顕著な成果をあげた。
1.神経伝達物質受容体の機能と分子構造 三品はNMDA受容体チャネルを構成するεサブユニットファミリーを発見し、その分子的多様性がNMDA受容体の機能的多様性の基盤となっていることを明らかにした。小澤はAMPA/カイニン酸受容体の脱感作の程度とkineticsがこの受容体のサブユニット構造によって規定されることを示した。赤池はGABA受容体の機能の研究の過程で、脳の特定の部位から特定のシナプス機能をもつニューロンの単離に成功し、シナプス研究のための新しい実験法を確立した。高橋はダリシン受容体の幼若型および成熟型cDNAの発現実験を行い、両受容体のチャネル機能の差異を明らかにするとともに、幼若型から成熟型への移行と脊髄におけるグリシン依存性の抑制性シナプス機能の発達との関連を明らかにした。杉山は代謝型グルタミン酸受容体の機能解析のための薬理学的ツールの開発を試み、ジハイドロキシフェニルグリシンがこの受容体に対する強力な選択的アンタゴニストであることを発見した。
2.細胞内Ca^<2+>測定法の改良と応用久場は共焦点レーザー顕微鏡を用いて、ニューロン内のCa^<2+>濃度分布を精細に測定しCa^<2+>誘起性Ca^<2+>遊離機構のもつ機能的意義を解明した。八尾は鶏胚毛様体神経節のシナプス前終末部の細胞内Ca^<2+>濃度の測定法を開発し、Ca^<2+>濃度変動と伝達物質放出との定量的関係を明らかにするための研究を開始した。立花は網膜双極細胞の軸索終末部における細胞内Ca^<2+>濃度の変動を定量的に解析し、Ca^<2+>チャネルを介して流入するCa^<2+>のうちわずか数百分の一が遊離状態にあることを算定した。工藤は海馬スライス培養標本で細胞内Ca^<2+>濃度の変動を指標としてグルタミン酸受容体サブタイプの発現の部位差を明らかにした。
