研究概要

シナプスの化学的情報伝達機構の研究で、特に次の三点において顕著な成果をあげた。
1.神経伝達物質受容体の機能と分子構造 三品はNMDA型グルタミン酸受容体を構成するεサブユニットファミリーのcDNAの単離に成功し、NMDA受容体の機能的多様性の分子的基盤を明らかにした。小澤はパッチクランプ法とPCR法を併用してCa^<2+>透過性AMPA型グルタミン酸受容体のサブユニット構成を単一ニューロンレベルで決定した。赤池は脳の特定部位からニューロンの単離を行い、GABA_b受容体の活性化が自発性抑制性シナプス後電流の発生頻度と電流量を減少させることを示した。高橋はグリシン受容体の幼若型および成熟型cDNAの発現実験を行い両受容体チャネルの特性に明瞭な差異が存在することを明らかにした。 2.細胞内Ca^<2+>の動態とニューロン機能 久場は共焦点レーザー顕微鏡を用いて交感神経節ニューロン内のCa^<2+>濃度分布を精細に測定し、Ca^<2+>誘起性Ca^<2+>遊離機構の存在することを示した。八尾は毛様体神経節のシナプス前終末からの伝達物質の放出にはN型Ca^<2+>チャネルの活性化が重要であるとともに、このチャネルはアデノシンにより抑制されることを明らかにした。立花は網膜双極細胞の伝達物質がグルタミン酸であることを証明するとともに、この伝達物質の放出にはL型Ca^<2+>チャネルの活性化が重要であることを明らかにした。工藤は海馬スライス培養標本で細胞内Ca^<2+>濃度の変動を指標として、グルタミン酸受容体サブタイプの発現の部位差を明らかにした。 3.シナプス機能の可塑性変化 平野は小脳培養系で顆粒細胞・プルキンエ細胞間のシナプス伝達の長期抑圧(LTD)の発現機構の解析を行い、代謝型グルタミン酸受容体mGluRlの活性化がLTDの発生に必須であることを示した。
