研究概要

全分担者において研究は申請時より着実に進展した。
1.毒性物質を同定したもの:歯周病菌に血球凝集素を精製した(吉村)。
2.遺伝子を同定、サブクロ-ン化したもの:腸球菌の溶血毒遺伝子(池)鶏病大腸菌のI型線毛遺伝子(関崎),H.pyloriのサイトトキシン遺伝子(中沢)。
3.構造遺伝子の一部の塩基配列を決定したもの:新しい下痢病原菌である腸管粘着性大腸菌の粘着因子遺伝子(山本・達),B.fragilisのノイラミニダ-ゼ遺伝子の一部を決定し、診断用プロ-ブを開発した(大西)。
4.構造遺伝子の全塩基配列を決定したもの:豚コレラ菌の細胞侵入性遺伝子群mba1〜4(檀原),緑膿菌のエラスタ-ゼ、アルカリプロテア-ゼ(奥田),ボツリヌスC型毒素遺伝子についてはさらに機能的ドメインも解明した(小熊)。
5.(4)のステップを完了し、調節遺伝子系その他の解明へ進んだもの:チフス菌V_i抗原の重合と輸送を支配する遺伝子(江崎),セラチアの潜在性線毛遺伝子の調節系(天児),コレラ菌溶血毒を活性化する遺伝子(山本・耕),腸炎ビブリオの耐熱性溶血毒の正の調節遺伝子tox‐R(西渕),緑膿菌のサイトトキシン溶原ファ-ジの宿主菌染色体への組込み機構(寺脇・林哲也),ウェルシュ菌Q毒素の活性化因子(林英生)などがこれに属する。
6.赤痢菌の細胞侵入性遺伝子群の解析:D群菌については浸透圧による調節がvirF遺伝子の発現調節を介して行われること(渡辺),B群菌については広汎な解析がなされており、宿主細胞への細胞侵入性を支配する遺伝子産物の一つが菌体内膜貫通性蛋白であること,および細胞間拡散を支配するvirG遺伝子を調節する遺伝子virKの塩基配列を決定した(笹川)。
7.その他:プラスミドRl00のTraT産物は補体成分C6のC5bへの結合を妨げることにより,菌の血清抵抗性に寄与することを証明した(井上)。
