研究概要

1.単純ヘルペスウイルス(HSV)の感染発症機構;1)HSVのCPE変異株の試験管内での選択において、補体存在下では非融合変異株が、インターフェロン存有下では融合変異株が選択された(新居)。2)Acyclovir耐性度が高く、チミジンキナーゼ活性の低いHSV-2臨床分離株で、マウスに対する病原性が低下していないYS4-C-1株について分子生物学的検討を行い、TK遺伝子の182番目のアミノ酸がセリンからアスパラギンに置換していることを見いだした(森)。3)PCR法で増幅した産物について、HSVの型別を決定する特異的primer pairとprobeを検討した(栗村)。4)5カ国のHSV分離株のRFLPの解析のため、制限酵素の切断部位の変動点の有無について検討した(坂岡)。5)プロテインキナーゼをコードするUS3領域を不活化したHSV-2変異株を作成し、親株と神経病原性を比較したところ、腹腔接種、後肢接種では1万倍以上低下していたが、角膜や脳内接種の場合は10倍程度の低下に留まった(西山)。6)HSV粒子に存在し、パルス電気泳動でHSVゲノムより泳動距離の長いDNAは、これまでに報告されている欠陥干渉ゲノムとは異なった構成を示すことが明らかになった(倉田)。
2.EBウイルス(EBV)の感染発症機構;1)原発性胃腺癌970例について、EBVゲノムの存在を検索した結果、67例(7%)が陽性であった。さらにEBVゲノム陽性細胞は上皮性癌細胞であり、これらはモノクロナールな増殖であることがゲノムの解析から明らかになった(大里)。2)潜在EBV活性化に働くスイッチ遺伝子BZLF1の上流のプロモータ領域を、10種のウイルス株について比較した結果、誘発刺激への応答に重要な領葵が、これらの株間で完全に保存されていた。またCAT assayによりプロモーター活性を比較したところ、Akataウイルスが特に誘発刺激への応答性が高いことが明らかになった(高田)。
3.サイトメガロウイルス(CMV)の感染発症機構;1)マウスCMVの温度感受性変異株とその復帰株、さらに原株と温度感受性変異株の組換え体を得て、新生マウスに対する病原性、病理像の比較を行った(南嶋)。2)ヒトCMYのgBの中和に関与するエピトープを解析するため、gBのN末端をpMal-C発現ベクターに挿入して、発現した融合蛋白を抗原として、患者血清との反応を検討した(古川)。3)MCMY潜伏感染マウスに誘導される抗唾液腺抗体(ASDA)とシェーグレン症候群の病因的関連を検討した(清水)。
4.ヒトヘルペスウイルス6(HHV-6)の感染発症機構;1)臨床材料よりHHV-6ゲノムを高感度に検出する方法として、Nested double PCR法の導入により感度を増大させた。またA型とB型の型別判定可能なPCR法が開発された(山西)。HHV-6および-7感染リンパ球の電子顕微鏡観察により、HHV-7に特微構造が認められた(新居)。
