研究概要

筋細胞のみならずあらゆる細胞には顕微鏡下では何らかの可視的な動きがみられる。この細胞運動一般は特殊な場合を除いてATPの化学エネルギーを“うごき"という機械的エネルギーに変換する蛋白質群によって担われている。これらはMotor蛋白質と呼ばれていて、アクチン・ミオシン系はこれに当たる。しかし、Motor蛋白質の試験管内での機能は単純であるため、これがどの様に多様な細胞運動を発現しているかという役割の問題にはほとんど手がけられていない。本研究ではこの役割を明らかにするために(1)新しいMotor蛋白質の検索、(2)アクチン・ミオシン系の機能を制御する新しい蛋白質の発見、(3)近年のアクチン・ミオシン系の研究に取り入れられて実績をあげつつある新しい技術(i,In Vitro Motility Assay;ii,蛋白質一次構造決定;iii,核磁気共鳴;iv,遠心顕微鏡法)の導入を目的としている。
3年間の研究計画の実施により(1)はいくつかの植物細胞よりミオシン様蛋白質の発見に、(2)は平滑筋細胞のミオシン軽鎖キナーゼの新しい機能の発見に結びつけることができた。(3)ではi)を平滑筋収縮制御の研究に、ii)を先体反応研究に応用することができた。さらにiii)はミオシンのmotor機能分析に、iv)はアクチンのmotor機能分析に応用することが出来た。
