研究概要

細胞活性化異常のメカニズムの研究はウイルス感染症が直接、関接に関係した発ガン機構、自己免疫的病態の解明に極めて重要である。本研究は、多面的な総合研究を行い、ウイルス疾患に関連した細胞活性化機構の基礎・臨床医学的研究を促進する事を目的とする。今年度得られた研究成果は、1)ADF/TRX遺伝子はウイルス疾患での異常発現が知られているが、今回ゲノムにおけるADF/TRX遺伝子の発現調節部位のクローニングを完全しSP-1,AP-1,HSEの存在を明かとした。さらにウイルス関連蛋白による制御を検討中である。また、ADF遺伝子を過剰発現した変異マウスを作製し,抗原提示能へのredoxの関与を明らかにした。さらにADF/TRX遺伝子を欠損したマウスを作製中である。(淀井)2)NFKBの前駆体とHTLV-1ウイルスのTAX蛋白が結合していることを明かとした。また、サイトカイン遺伝子特に、GM-CSF遺伝子上流の転写調節について詳細に検討し、調節機構を明かとした。(新井)3)SCIDマウスにHTLV-1細胞株あるいはATL患者リンパ球を導入し、ATL発症動物モデルの作製に成功した(内山)。4)HAM患者髄液および血清中の各種サイトカイン測定を行い、病態との相関を明らかとした、また,研究代表者との共同研究でretinoic acidがHAM患者リンパ球の自律的増殖抑制に効果的であることを示した。(納)。5)慢性関節リウマチ(RA)などの病的組織に、IL-6またはIL-6R遺伝子発現を誘導しうる因子の解析を継続している。(平野)6)慢性疲労性症候群におけるウイルスの関与および、NK活性異常の検討を行った。(内田)7)どん食などのマクロファージ活性化によってADF/TRXが誘導されることを明かとした。(淀井、翠川)8)ウイルス疾患での免疫異常を解析するため、新たなIL-1受容体関連分子をクローニングすることを成功した。(寺田)ひきつづきこれらの成果を発展させ、ウイルス疾患での免疫細胞活性化異常の研究を有機的に進めたい。
