研究概要	プラスチドは光合成お物質生産を担う植物特有のオネガネラで独自のゲノムと完結した遺伝情報発現系をもつ。プラスチド遺伝子の発現制御は主として転写後(RNAレベル)で行われる。そこで、我々は転写後調節に関与すると考えられるRNA結合タンパク質を単離し、その遺伝子の構造と発現様式および機能の解析を進め、初年度は以下の成果を得た。 1)タバコ葉緑体の3種類の葉緑体RNA結合タンパク質(リボヌクレオプロテイン)cp28、cp31、cp33を無細胞RNAータンパク質合成系で合成し、それぞれの核酸歩合能を調べた。3種のタンパク質とも1本鎖DNAと強く結合するが、特にポリ(G)とポリ(U)と高い結合特異性を示した。 2)RNA結合タンバク質cp31の遺伝子を単離して、その全塩基配列を決定した。この遺伝子は4個のエキソンと3個のイントロンよりなり、転写開始後は翻訳開始点より168bp上流に存在する。転写産物の中にはオルタナティブ・スプライシングによるものは含まれていない。Nicotiana sylvestrisの核ゲノムにはこの遺伝子は1コピ-存在することが明らかとなった。 3)新たに2種の葉緑体RNA結合タンパク質cp29Aとcp29Bを同定し、各々のcDNAと遺伝子を単離・解析した。両者ともトランジト・ペプチド、酸性ドメイン、2個のRNA結合ドメインを持っていた。また両遺伝子は4個のエキソンと3個のイントロンより構成されていた。 4)以上5種のcpタンパク質に存在する10種のRNA結合ドメインの構造が明らかになったので、その系統樹を作製した。
