研究概要	プラスチドの遺伝情報発現系を構成する因子の多くは核ゲノムにコードされている。我々は、転写後過程に関与するRNA結合タンパク質を単離し、その遺伝子の構造と発現様式及び機能を解析した。 1.タバコ葉緑体RNA結合タンパク質cp29A遺伝子を単離して全塩基配列を決定した。この遺伝子は749bp,258bp,303bpの3個のイントロンを含み、これを除いたリーデングフレームは291コドンより成っている。転写開始点は近接する2ヶ所よりなり、それぞれ前方にTATAボックス様配列が存在する。 2.葉緑体RNA結合タンパク質は1個の酸性ドメインと2個のRNA結合ドメインよりなる。その核酸結合能を調べるため、cp29AとBより各種の欠失変異蛋白を作製し、それぞれの核酸結合能を測定した。いずれも、酸性ドメインを除くと、元の蛋白に比べ結合能が高まった。RNA結合ドメインは1個でも元の蛋白(2個)に比べ弱いがポリGとポリUに結合することから、このドメイン1個で、RNA結合性を持つと考えられる。スペーサーはあってもなくてもRNA結合ドメイン1個の結合能に関係しない。しかし、RNA結合ドメイン2個を結合した蛋白(スペーサーがない)も1個のドメインと同じ結合能であった。このことはスペーサーがRNA結合ドメインの結合能の加算効果に必須であることを示す。 3.葉緑体伸長因子FF-Tuの2種の遺伝子(tufAとtufB)は、異なった転写様式を示すことを明らかにした。また、葉緑体リボソーム蛋白質についてもL28のcDNAを単離・解析した。
