研究概要

1.新しいRNA結合蛋白cDNA:昨年度のRNA結合蛋白RGP-1につづいて新たにRGP-2のcDNAを単離した。RGP-2蛋白は、N-末ドメイン(39アミノ酸)、RNA結合ドメイン(82アミノ酸)、グリシン・リッチ・ドメイン(83アミノ酸)とC-末ドメイン(46アミノ酸)よりなる。葉緑体への輸送実験よりRPG-2は核・細胞質系に存在すると考えられる。この蛋白はポリ(G)とポリ(U)に高い親和性を示す。
2.プラスチドRNAへの結合:5種のプラスチドRNA結合蛋白は成熟tRNAを除くプラスチドRNAへ結合した。従って、これらの蛋白はプラスチドDNAよりRNA転写が起きるとそれに結合し、RNAの保存やリボゾームへのmRNAの受渡しに関与すると示唆される。
3.プラスチド翻訳系:効率的なin vitro翻訳系の開発に成功したので、この系より5種のRNA結合蛋白を抗体沈降により除き、psbAのmRNAの翻訳を調べたところ、翻訳の促進がみられた。このことはRNA結合蛋白が翻訳前のmRNAの保護に関与していると考えられる。
4.プラスチドのRNAエディティング:数種のタバコプラスチドmRNAがRNAエディティングされることを見出した。プラスチドより調製したin vitro転写系およびin vitro翻訳系に前駆体mRNAを入れエディティングが起きる条件検討をしたが、現在までエディティングが起きる系は開発されていない。
