SCIDマウスを用いた単純ヘルペスウイルスの潜伏及び感染防御機構の研究

研究概要

免疫不全状態の宿主における単純ヘルペスウイルス感染の発症病理の解明を行った。免疫不全状態の動物モデルとして、SCID(重症複合型免疫不全)マウスを用いた実験に加えて、T細胞機能を欠損しているヌードマウスも使用した。感染防御及び免疫病理学的機序への関与の考えられる、種々の免疫担当細胞サブセットの細胞自動解析装置(FACScan,当補助金にて購入)を用いた解析法を確立した。マウスやヒトの血液、脾、リンパ節の他、角膜、網膜など炎症の実際に起こっている組織について、種々の細胞集団の検討を行った。マウスリンパ球サブセット表面マーカーに対するモノクローナル抗体を用いた、特定のサブセットの除去については、生体及び試験管内のいずれについても手法を確立し、解析に使用した。またDNA合成装置(当補助金にて購入)にて作成したオリゴヌクレオチドをプライマーとしたpolymerase chain reaction(PCR)法ならびRNA PCR法も手技を確立した。そしてウイルス感染細胞、実験感染動物の諸臓器における、単純ヘルペスウイルス由来DNAおよびRNAの検出ならびに解析を進めた。潜伏および再活性化の機構解析のためにいくつかのウイルス遺伝子の転写レベルの経時的半定量的検出を行った。再活性化において調べた遺伝子のうち前初期遺伝子のひとつ、ICPOの転写開始のみが検出可能な場合と、その他の遺伝子の転写産物も検出可能な段階まで起こる場合のあることを報告した。また特異な病原性を示した単純ヘルペスウイルス分離株について、その病原性に深く関与していると思われる遺伝子(チミジンキナーゼ遺伝子及びエンベロープ糖蛋白C遺伝子)を中心に分子生物学的解析ならびにマウスにおける発症病理の解析を行い、報告した。さらにPCRによる唾液中の単純ヘルペスウイルスDNA検出と従来のウイルス分離との比較検討を行い、報告した。
