骨形成因子の合成とその臨床応用のための基礎的研究

研究概要

骨形成因子は、正常の骨形成細胞(骨芽細胞)によって産生される骨の微量成分をなす生理活性タンパク質である。その生理活性によって人為的に体の任意の部分に骨形成を誘発できる。この因子は臨床に有効に応用すれば骨折の治癒促進や欠損修復、人工骨と骨との接合等に有効であろう。本研究は、この骨形成因子の応用のための生理的制御機序の解明等を目ざすものである。
本年度までに完了した成果について以下に記す。
1)高等動物での骨形成因子drug delivery systemの確立。
骨形成因子の活性発現には適当な担体を使ったDrug delivery systemが必要である。また、霊長類(ヒト)では、骨形成因子は作用しにくいことも知られており、さらに適切なDDSが必要である。本研究の結果、1)適当な特性をもった担体として、合成高分子ポリマーであるポリ乳酸ポリエチレンブロック重合体を開発し、2)この担体と骨形成因子を複合し、さらに多孔性セラミックスと複合することで霊長類に再現性よく骨形成を誘発できることを明らかにした。すなわち骨形成因子の臨床応用の可能性を示した。
骨折修復反応での骨形成因子の役割
骨折修復での骨形成因子の役割りを明らかにするための骨折修復組織内での骨形成因子の遺伝子発現をマウスを用いて検索した。その結果、骨形成因子は修復反応の初期(骨折後1〜2日)に骨折部近傍のやや未分化な骨膜内の細胞や骨髄内の未分化な細胞に一過性に遺伝子発現の亢進がみられることを明らかにした。
3)骨形成因子の阻害因子の検索
骨形成因子の作用制御機序の手がかりをつかむために骨形成因子の阻害因子の検察をした。多くの生活活性物質の骨形成因子の制御効果をスクリーニングした。その結果炎症性サイトカインであるTEN-αが骨形成因子の制御効果を確認した。その作用機序について研究を行っている。
