時間分解蛍光装置と共焦点顕微鏡を用いる細胞のエンドサイト-シスの研究

研究概要	真核細胞のエンドサイト-シスの分子機構解明を目的とし、培養細胞中における「単一の」エンドソ-ムの膜融合を、顕微鏡下において定量的に測定した。融合アッセイには、2種の蛍光色素(エネルギ-供与体;Dとエネルギ-受容体;A)それぞれでラベルしたタンパク質リガンド(α_2マクログロブリンなど)を含むエンドソ-ムの融合に伴う内水相混合の結果、DーA間の蛍光共鳴エネルギ-移動(RET)効率が増加し、Dの蛍光寿命が短くなることを利用した。蛍光寿命の見積りには、当研究室で開発した時間分解顕微蛍光装置により、顕微鏡下、直径約1.5μmの微小領域内にある蛍光リガンドの入った「単一の」エンドソ-ムからのDの蛍光減衰曲線を精度良く、短時間(約60秒)で測定し、その減衰曲線よりDの蛍光寿命を見積った。その結果、単一のエンドソ-ムの融合がNH/3T3細胞内で確かに測定でき、エンドソ-ムの融合は、エンドソ-ム内の酸性化や、細胞骨格であるアクチンフィラメントには非依存的であるが、微小管には部分的に依存していることを明らかにした。しかし、MDCK細胞のような厚みのある細胞内でのエンドソ-ム融合の程度の空間的分布を見るには、z軸(深さ)方向から来る他のエンドソ-ムからの迷光や、細胞の自家蛍光の為に、個々の減衰曲線の測定精度がどうしても低下する。現有の時間分解顕微蛍光装置の顕微鏡部を共焦点レ-ザ-顕微鏡にすることで、従来の光学顕微鏡において達成しにくかったz軸方向の空間分解能を飛躍的に向上させうる。その為、現在、蛍光励起用のパルスレ-ザ-光(繰り返し頻度4MHz、波長365nm、パルス外部幅約8psec)をできるだけ強度を保ったまま外部レ-ザ-として共焦点レ-ザ-顕微鏡に導入し、試料からの蛍光をシンクロスキャンストリ-クカメラよりなる検光部で感度良く検出するための中間レンズ、フィルタ-系の改良を行っている。
