新しい脳下垂体ホルモン-ソマトラクチンの機能と遺伝子の研究

研究概要	ニジマス脳下垂体中葉におけるソマトラクチン(SL)の発現と細胞内局在をシロサケのSLcDNAを用いて、インサイチュ-ハイブリザイゼイション法で証明した。また、金コロイド法によりSL免疫活性が分泌顆粒中にあることを示した。これらの細胞学的所見はSL細胞が内分泌細胞であることを支持する。昨年度までに、性成熟、環境水のカルシウム濃度の変動にともなって血中SLレベルが変動することから、SLがホルモンと定義できる要件を満たした。SLの機能を特定する研究の一環として、ニジマスを酸性水中に移したところ、血中のGHとPRLレベルの変動は認められないが、SL濃度が上昇することを発見した。この結果は、SLが酸・塩基平衡の調節に関与することを強く示唆している。シロサケSLのゲノム遺伝子の転写開始点の上流域に、哺乳類のGHおよびPRL遺伝子の転写因子であるPit-1の結合配列を見出した。シロサケのPit-1遺伝子を魚類で初めて同定した。365アミノ酸残基からなるシロサケPit-1に特異的な構造部分を含む融合タンパク質を大腸菌で発現させ、これを抗原として抗体を作成した。免疫組織染色により、ニジマスの脳下垂体の前葉のGHおよびPRL細胞の核のみならず、中葉のSL細胞の核にもPit-1が局在することを証明した。次に、Pit-1遺伝子を発現ベクターの下流に挿入したプラスミドとSLおよびGH遺伝子の転写調節領域をルシフェラーゼ遺伝子の上流に挿入したプラスミドをヒーラ細胞に導入し、ルシフェラーゼ活性の増大を確認した。即ち、Pit-1は、GHのみならずSL遺伝子の転写調節領域に作用して、転写を活性化すると解される。このSL遺伝子のPit-1による活性化に必要な5′上流領域のサイズは0.5kbであった。
