タ-ゲティング型製剤の製剤特性ー機能相関の体系化

研究概要

本研究では、薬物の体内分布過程を制御し特定の標的部位に送達させると共に作用部位以外への臓器への移行を回避することにより、薬物適用と治療の最適化を図る“薬物タ-ゲティング"の学問および技術体系の確立を目的として、(1)標的指向化効率の評価解析法の確立、(2)運搬体システムの体内挙動特性の系統的整理、(3)標的指向化の方法論の確立、の三つの観点より検討を行った。第一の問題については、標的指向化に利用される各種運搬体システムの全身レベルでの動態特性を、生理学的薬物速度論を基礎とし解析する方法を確立し、引き続いて臓器潅流実験法や細胞培養系を利用して臓器、細胞レベルにおけるこれらの動態機構を全身レベルの動態と関連づけて解析する実験法を開発した。こうした実験系を利用して、各種高分子性あるいは微粒子性運搬体システムの動態特性をそれぞれの物理化学的あるいは生物学的特性と関連づけて整理した結果、前者に関しては分子サイズや電荷などの物理化学的特性あるいは糖鎖構造などの生物学的機能性官能基が肝臓への取り込み、腎排泄あるいは癌病巣などの標的臓器への移行に及ぼす影響を明らかにすることができた。また、各種微粒子性運搬体の製剤特性と動態の関連についても同様にいくつかの新しい知見を得た。そこで、以上のような知見を基礎として確立した設計指針に基づき実際に抗癌剤などを対象薬物としてその応用の可能性を検討した結果、モノクロ-ナル抗体を利用した癌病巣への送達システムや、糖鎖認識機構を利用した肝臓実質細胞への送達システムの開発に到達し、こうした体系化された情報に基づく総合的設計アプロ-チの有用性が確認された。
