微小管・細胞膜架橋蛋白質に対するジベレリンの影響

研究概要

1.タバコ培養細胞からの微小管架橋蛋白質の単離
タバコ培養細胞より調製したプロトプラストをパーコル上に重層し遠心すると、液胞を持たない、いわゆる、ミニプロトプラストを得る事ができる。このミニプロトプラストから得た抽出液中のチューブリンが、微小管安定化剤の存在しない条件下でも重合をすること、また、重合した微小管は架橋構造を介して、束を形成していることを明らかにした。さらに、チューブリンを重合・脱重合法で純化した際にチューブリンと共に、140Kd,110Kd,65Kdの蛋白質が純化されることを見いだし、このうちの65Kd蛋白質を、陰イオン交換クロマトグラフィーにより単離し、この蛋白質に微小管を束化する働きがあることを確かめた。また、抗体を調製し、間接蛍光抗体染色法により、65Kd蛋白質が細胞内でも微小管と同じ位置に存在していることを明らかにした。
2.タバコ培養細胞の微小管・細胞膜架橋構造
1991年度の研究により、タバコ培養細胞より調製した膜ゴーストが、タバコ培養細胞のミニプロトプラストの抽出液中で重合した微小管を付着させること、このゴーストの微小管付着能が、ゴーストをプロテアーゼ、または0.6M KC1溶液で処理することにより取り除かれることを明らかにしたが、ゴーストをKC1処理した場合でも抽出液にタキソールを加え、重合を促進、あるいは微小管を安定化させると付着が起きることを見いだし、KC1でゴーストより取り除かれる因子が微小管を安定化させる因子である可能性が高いことを示した。また、粗抽出液中で重合させた微小管はKC1処理したゴーストに付着するが、精製したチューブリンを重合させた場合には、タキソール存在下でも付着しないことを明らかにし、粗抽出液中に微小管を細胞膜に付着させるのに必要な因子が含まれている可能性が高いことを示した。
