新規わい性発現物質A-2α、βの化学構造の解明と園芸への応用

研究概要

初年度に検討した大量抽出精製法に従いわい性エンドウ地上先端3節の節間および葉30kgを処理し精製を進めた。抽出、中性画分および第一段階のKieselgelカラムクロマトグラフまでは初年度の成績のとおりの収率で精製が進行した。しかし第2段階のKieselgelカラムクロマト(および/又はその後の濃縮、貯蔵中)においてA-2はほぼ完全に失活した。この原因は今後追及しなければならない。
抽出・精製の過程ではクレステスト(生物検定)によって活性物質の検定、定量を行うが、この他に高速液体クトマトグラフィー(HPLC)の併用により行程所要時間の短縮と定量精度の向上を計るため順相と逆相の2段階のHPLC法の確立を試みたが、試験した順相カラムではすべて活性物質の回収率が悪く成功しなかったので、Merk社のLiChroprep S60をつめたオープンカラムを第一段階としてWaters社の逆相Sep-Pak tC_<18>で前処理して、逆相HPLCカラム(TSKgel ODS-120T)で分析できる見通しを得た。
これらの過程でA-2画分に混在していたA-2とは異なる抑制物質A-1-3αおよびβを少量ながら単離し、ガスクロマトグラフ(GC)上のピークを特定することに成功した。またガスクロマト-マススぺクトル(GC-MS)測定の結果、分子量はともに250であるフラグメンテイションパターンからαとβは幾何異性体であり、溶液中で相互変換することを明らかにした。
