葉の老化に伴う窒素転流の分子的基盤-葉緑体プロテアーゼによるRubiscoの分解

研究概要

本年度はRubiscoのLarge subunit(LSU)の分解物の検索とその特性について明らかにすることとした。
コムギ葉を材料に、葉緑体を単離精製し、LSUの分解物について調ベた。サーモリシン処理により葉緑体の外包膜に付着しているタンパク質を除いたあとの葉緑体にも、LSUの分解物がRubiscoのそれに対し0.1〜0.2%のオーダーで存在していることが精製抗LSU抗体を用いた実験により示された。またこのことは、LSUのアミノ酸配列の中央部の15アミノ酸残基を含む合成ペプチドに対する抗体を用いた実験からも確かめられた。さらにN-末端およびC-末端近傍のアミノ酸15残基を含むペプチドを合成し、それらに対する抗体を調製し分解物との反応性について検討した。その結果、LSUの53KDaに対し、50〜40KDaにそれぞれの抗体に反応性のあるペプチドが複数存在することが分かった。つぎに葉緑体のストローマ画分をゲル濾過に供し、分解物がどの画分に見い出されるかを調ベたところRubiscoと同じ画分に含まれていることが示され、Rubiscoが4次構造を保ったままでN末端,C末端から分解を受けている可能性が示された。
また、これらの研究の過程で、LSUより高分子側に抗LSU抗体と反応性のあるポリペプチドが多数見いだされ、その量はLSUに対し、0.1〜0.2%前後であった。これらが実験の過程で人工的に生じた産物なのか、in vivoでのRubisco分解に対し積極的な意味を持つのか、現在検討中である。
