卵巣機能を調節する新しい生理活性物質群の単離・同定

研究概要	平成4年度において卵巣機能を調節する2つの生理活性物質を分離しその性状を解析したが、詳細は以下の通りである。 (1)卵核胞崩壊を抑制する卵母細胞膜因子:トリプシン溶液で0.5〜1時間処理したホツキ貝の卵母細胞は卵核胞の崩壊を誘起した。グリコシダーゼ DNase RNase処理は卵核胞崩壊に影響しなかった。トリプシン処理による卵核胞崩壊誘起にはCacl2が必要であり、その最適濃度は20mMであった。卵母細胞を1M尿素、5mMEDTAを含む10mMのトリス緩衝液で30分間培養して得た細胞膜因子は0.4〜0.8mg/mlの濃度でトリプシンによって誘起される卵核胞の崩壊を抑制した。当該因子を同定するために、抽出卵母細胞膜成分をセファデックスG100のカラムで分画したところ、分子量18KD付近に溶出する画分に強い活性を認めた。部分的に精製された因子は0.1〜0.2mg/mlで活性を示した。以上の結果はホツキ貝の卵核胞の崩壊が卵母細胞の膜成分によって抑制されており、カルシウムイオンの存在下でプロテアーセが卵母細胞の膜因子を分解して抑制を解除し、卵核胞崩壊を誘起するという考えを導びき出すものである。 (2)黄体で特異的に発現するコラーゲナーゼインヒビター遺伝子の同定:顆粒膜細胞や黄体細胞で発現する生理活性物質を系統的に分離するために顆粒膜細胞と黄体細胞のcDNAライブラリーを作製し、ディファレンシャルスクリーニングを行ない、顆粒膜細胞と黄体細胞で特異的に発現のみられるクローンを分離した。黄体細胞で特異的に発現のみられた遺伝子を解析したところ、コラーゲナーゼインヒビターをコードする遺伝子であることが明らかにされた。免疫組織化学的な解析を行ない、黄体の血管系の分岐とコラーゲナーゼインヒビターの発現に相関のあることを認めている。
