神経内分泌系における側性制御調節機構の分子細胞化学的アプローチ

研究概要	本年度の研究では、視床下部への入出力様相について免疫組織化学法とin situハイブリダイゼーション法を主に用いて検索した。まず、脊髄への入力機構を調べるために、現在最も高感度逆行性標識物質として注目を集めているコレラトキシンの脊髄への取り込みを検討した。次に、脊髄前後根切断によって、脊髄内神経伝達物質がどのような変動を呈するのか、経時的に追究した。あわせて、胎児期の脊髄を動物の前眼房内に移植し、神経伝達物質の局在について観察した。これらの実験から、脊髄内に存在する伝達物質は可塑的変動を行うものの、成熟時の分布をその後も保持することが明かとなった。動物の卵巣を摘出したのち、エストロゲンを長期間投与し続けると、視床下部のとくに前部の視束前野では、著しい神経線維の増加を示した。それらは、おもにCGRPやエンケファリンをその伝達物質として用いる系であることが免疫組織化学法やin situハイブリダイゼーション法により証明された。逆に、エストロゲン投与によってドーパミン系の神経線維はその数を減じた。そして、これらのニューロンがともにエストロゲン受容体を有することを明らかにした。これらの変化がin vitroでも生じるのか分散培養系の確立につとめた。視床下部からのニューロンはエストロゲンによく反応し、神経突起伸長とともに細胞骨格蛋白の発現も認められた。以上から、視床下部領域への入力や出力は、脊髄を介して側性性に制御されるものと、体液性の受容体を介して非側性性に統御されるものがインテグレイトされた総和として機能していることが明かとなった。
