レトロウイルス外膜蛋白活性化プロテーゼの発見と、その活性化による感染機序の解明

研究概要

癌ウイルス,エイズウイルスを始めとするレトロウイルスの外膜糖蛋白は、いずれもリボソームで前駆体として合成された後、宿主細胞のゴルジ膜で限定分解を受け小胞体ーゴルジ装置系を通って細胞膜へ選択的に運ばれることが知られている。この限定分解が阻止されると、ウイルスは不完全ウイルスとなり感染性を失う。我々はCD4陽性人Tリンパ球の膜分画より、ゴルジ膜あるいはERに局在するHIV-1の外膜蛋白前駆体活性化プロテアーゼの精製に成功した。本酵素は、HIV-1外膜蛋白前駆体gp160をgp120gとgp41に変換する分子量26kDaのトリプシン型セリン性プロテアーゼで、活性基のセリンを持つ。本酵素はgp160を基質にすると、pH6.5-7.5に至適pHを示した。本酵素はgp160のシークエンスの中で、-Arg^<508>-Leu-Lys-Arg^<511>-を特異的認識してArg^<511>の後を選択的に切断した。この切断部位のシークエンス-Arg(Lys)-X-Arg(Lys)-Argは、殆ど全てのレトロウイルス外膜糖蛋白前駆体の切断部位に認められる共通構造である。本酵素の活性は、人やマウスのT細胞以外に、B細胞、単球、HeLa細胞、血管内皮細胞に認められ種々のレトロウイルスの感染に関与していると思われる。本酵素の欠損した人マクロファージ培養細胞U2ME7ではHIVの外膜蛋白gp160は分解されずER、ゴルジに蓄積される。本酵素は、これまでに報告のあったDibasicの基質を特異的に分解するKEX2ファミリーのプロテアーゼと異なり、金属非要求性でEDTAは活性に影響を与えない。また本酵素の活性は、DFP、アプロチニンなどのトリプシンインヒビターとDTT、システインなどのSH試薬により抑制され、これらの中でも特に低分子インヒビターが培養細胞レベルでも有効であり抗HIV活性を示すことが明らかとなった。
