単クロ-ン抗体応用による腎障害機序の分子レベルにおける解析

研究概要

1.蛋白尿惹起単クロ-ン抗体(MA)5ー1ー6、1ー22ー3の大量精製:従来腹水化に難渋していたMA5ー1ー6につき、BALB/C腹腔内にて強い増殖を示したクロ-ンを引続きマウス腹腔内に植え継ぐことにより大量作製が容易となった。2.両MA惹起病変に及ぼす補体の影響:Cobra Venum Factorの応用によりMA5ー1ー6は補体非依存性に、MA1ー22ー3は補体依存性に腎病変を惹起することが確認された。3.^<125>I標識MAによる定量的検索:両MAとも大変微量(各々注射量として125μg,25μg、両腎結合量として12.8μg,1.2μg)にて腎病変を惹起することが明かとなった。これは従来の実験腎炎における病変惹起必要最低量に比較して桁違いに微量であり、余分の免疫反応が除外され得るMAならこその値であり、かついかに限定された重要部位との反応が引金を引くことになるかを示すものである。4.腎糸球体可溶化による対応抗原の同定、並びに性状検索:従来の常套手段を繰り返し駆使して鋭意努力中であるが難航している。対応策として(1)抗原の絶対量が大変少量である可能性を勘案してより大量の腎組織を出発材料とした。(2)ELISAによる対応抗原検索法を確立した。(3)MAと同一の特異性を有するウサギポリクロ-ナル抗体の作製を開始した。(3)の試みはcDNAライブラリ-からのMAによるクロ-ニングの難しさに関するウイ-ン大学教授Kerjaschki博士からの体験に基づいた示唆に従ったものである。5.対応抗原の動態検索:MA静注後、対応抗原は腎局所において減少、消失し、1日後には小胞体に証明された。補充のための生合成像と解されている.6.両MAの反復投与・複数投与による新モデルの開発:MA1ー22ー3反復投与により進行生の硬化性病変が得られつつある。これは慢性化機序解明という最重要課題に有力な武器を提供することになるものと期待される。
