クロイツフェルト・ヤコブ病プリオン蛋白のシナプス内局在の同定

研究概要

クロイツフェルト・ヤコブ病(以下CJDと略す)の原因物質としてプリオン蛋白が提唱されている。以前までは、組識学的に異常プリオン蛋白はクル斑と呼ばれるアミロイド沈着にのみ証明可能であり、CJD患者の大部分では、異常プリオン蛋白の局在を同定しえなかった。今年度我々は、加水分解の原理に基づいた前処理法を開発し、CJD患者全例に異常プリオン蛋白が証明可能であることを報告した。検出できた異常プリオン蛋白は中枢神経系の灰白質に広く分布し、シナプスを染色するシナプトフィジン抗体の分布と同一であった。さらに詳しく2重染色にてシナプスとプリオンの免疫染色性が一致することを同定した。またCJD患者脳より生化学的にシナプトゾ-ム、ミクロゾ-ム、シナプス小胞を分離し、シナプトゾ-ムに最も高濃度にプリオン蛋白が存在することを証明した。さらにプリオン蛋白遣伝子に異常をもつ患者でプリオン蛋白の分布を検討すると、シナプスにプリオン蛋白が沈着するのはコドン200の点変異を有する症例でみられ、コドン102、コドン117、コドン129の点変異や168bpのインサ-ションを有する症例では、ほとんどシナプスには沈着せず、細胞外のアミロイド斑として沈着するのが主であることを証明した。アミロイド斑の存在する症例では、臨床経過が非常に長く、シナプスが障害された症例では、ごく短い臨床経過であることが明らかとなり、異常プリオン蛋白の沈着形式が直接臨床病型に影響をおよぼす可能性が示唆された。さらに今年度開発した加水分解法をCJD感染マウスに応用することにより、脾臓やリンパ節の樹状細胞に異常プリオン蛋白が蓄積することを明らかとした。今年度我々が明らかとしたプリオン蛋白の局在は、従来我々が報告したクル斑以外に、新たにシナプスと樹状細胞が加った。
