炎症局所で見いだしたIL-1抑制因子の構造と機能

研究概要	ウサギ腹腔でのIL-1産生の動態を、その生物活性、抗原の両者で測定すると、その出現の時間に両パラメーターの間に食い違いが認められ、抗原の発現は、活性の発現より著しく長く持続していた。この原因の1つとして、炎症後期にIL-1抑制因子が産生されていることを見出し、これを分離、精製し、その1次構造を決定した。この因子は等電点5.5、分子量16kDのペプチド因子で、IL-1との結合、分解能はなく、タイプI、タイプIIいずれのIL-1レセプターへのIL-1α、βのいずれのリガンドの結合も阻止する因子であった。この炎症局所のIL-1抑制因子は、そのN端がブロックされていたため、リシルエンドペプチダーゼフラグメントを作製し、5種のフラグメントにつき、そのアミノ酸配列を決定し、N端がブロックされていたフラグメントのアミノ酸分析を実施した。これらの判明した1次構造に基づき、2種のオリゴDNAを合成し、PCR法により400bpのプローブを作製し、ウサギ腹腔の24時間滲出細胞から作製したcDNAライブラリーを検索し、陽性クローンの選択とその構造の検索を実施した。一方、N端がブロックされていたフラグメントのアミノ酸分析値、マススペクトルから、N端の構造とその修飾分子を決定し、この分子がアミノ酸143ケから成る因子で、ヒトのIL-1レセプターアンタゴニスト(IL-1ra)に相当する分子であることを決定し、遺伝子組換体、抗体の作成と、免疫測定系を開発し、ウサギの炎症におけるこの因子の役割を追求するシステムを完成し、膝関節炎をモデルとして検索を実施した。その結果、IL-1raは炎症早期から、後期にわたって、モル比でリガンドの200倍産生されており、炎症反応を制御していること、組換体を外から与えると、炎症反応、及び炎症に伴う組織破壊、いずれも阻止できることを明らかにした。
