ヒト胚細胞腫瘍細胞の分化能に関する分子細胞生物学的研究ーヒト体細胞分化の実験的アプローチー

研究概要	新たに樹立したヒト胎児性癌細胞(EC細胞)NCR-G2,G3細胞を用いて,以下の研究を行なった。1)両細胞の分化能をさらに詳細にする。2)多分化能を有するG3細胞の分化初期に関連する遺伝子をサブトラクション法により単離し,その機能を明らかにする。3)多分化能G3細胞からその分化・増殖に関わる機能的分子を確立する。その結果,1)Retinoic acid(RA)投与によって分化形質の発現が認められなかったG2細胞はhexamethylene-bis-acetamide(HMBA)の投与によって細胞骨格蛋白特に,神経線維蛋白の発現が蛋白およびmRNAレベルで認められた。ヒトEC細胞の分化の方向性は潜在的に有する分化能とともに分化誘導物質の性質によっても決定されることが明らかになった。2)多分化能を示すG3細胞のRAによる分化初期に関連する遺伝子のうち,90kD熱ショック蛋白遺伝子(HSP90β)の機能を解析した。RA添加後2時間後からmRNAレベルでHSP90の発現が増強した。その後10日間でG3細胞は栄養膜細胞への分化が認められた。その際,totalの蛋白レベルではmRNAに相当する発現量の変動はなかったが,フローサイトメーターでの細胞レベルの解析ではRAまたは熱ショックによりHSP90低発現群細胞の増加と高発現群細胞の減少および発現増強という結果が得られた。G3細胞の分化においてHSP90のdownまたはupregulationのどちらが機能しているかを明らかにする系を確立しつつある。3)G3細胞を免疫原としたモノクローナル抗体6E2はヒトEC細胞および正常胚細胞の細胞表面と特異的に反応し80kD蛋白を認識する。さらに,同抗体は量特異的にEC細胞の死を誘導し得ることが明らかとなった。この機序を解明することおよび本分子の生化学的性状を詳細にすることが今後の課題である。
